宋美龄1899年3月30号（光绪23年三月初五）在上海出生。父亲是卫理公会牧师宋嘉树，被堂舅收为儿子后，带到波士顿。宋家三姊妹因而得以在美国开放以及中国保守的环境下成长，并主导中国近代史的演变。
大姐蔼龄1904年只身赴美就读Wellesley College，为两个妹妹作开路先锋。之后1907年，十岁的美龄与十四岁的庆龄一同赴美念书。1912年，15岁的美龄成为Wellesley的正式生，选读英国文学、哲学及法文等。在这学校时，就显出她的领导才能。
宋美龄的母校，Wellesley College就在Boston的西南，这所声名显赫的女子学院知名校友除了宋氏三姐妹外，也包括希拉里·克林顿和美国前国务卿奥尔布莱特。

在进入之前，三姐妹先在乔治亚州的另一所女子学院Wesleyan College学习过，这也是美国最早的女子学院。1943年，该校授予三人荣誉博士学位。
